In what was a fiery clash, Uruguay ended the match with nine men after both Edinson Cavani and Jorge Fucile were sent off as Chile booked a date with either Peru or Bolivia in the semi-finals.
The opening half was dominated by Chile, with Alexis Sanchez, Jorge Valdivia and Arturo Vidal all coming close to opening the scoring.
Vidal sent a scare through the Chile camp after he limped off the ground following a strong challenge, but returned for the second half.
The clash had plenty of needle, and Uruguay star Edinson Cavani was given his marching orders after receiving a second yellow card for a perceived headbutt on Chile defender Gonzalo Jara.
Replays showed Jara had provoked Cavani by putting his finger in towards the Paris Saint-Germain attacker’s anus, leading the Uruguay man to retaliate.
There wasn’t much contact, but Jara made the most of it and Cavani exited the match in the 68th minute.
From then on Chile piled on the pressure, and the match-winner came from the most unlikliest of sources with defender Isla cooly finishing from the edge of the box with less than 10 minutes remaining.
