Valencia are believed to be desperate to keep hold of the Argentine and knocked back a transfer bid from the Red Devils in January.
According to Metro however, United will try their luck again with Otamendi believed to be wanting a move to the Premier League.
The 27-year-old still has four years remaining on his current deal with the La Liga side and United will have to dig deep into its coffers to sign the former FC Porto star.
Otamendi could be the fix to Louis van Gaal’s issues in central defence after the Dutchman failed to settle on a solid combination this season.
Chelsea has previously been interested in Otamendi, but it now seems the Red Devils are the front-runners to snare the Argentine international.
Otamendi has steered clear of speaking publicly about the proposed move, saying he would prefer to concentrate in helping guide Valencia into the Champions League.
Valencia currently sit fourth on the La Liga table, three points clear of fifth-placed Sevilla.
