The Spaniard finished fifth in the season-opening Grand Prix of Qatar, but he bounced back in Austin.
Marquez captured pole position in dramatic circumstances and was dominant in the race, finishing 2.3 seconds ahead of Ducati rider Andrea Dovizioso.
Nine-time world champion Valentino Rossi rounded out the podium, crossing the finish line in third on his Yamaha.
The race was delayed just moments before the start due to water patches being spotted underneath the footbridge at turn three, however it was Dovizioso who got off to the best start, leading into turn one.
With the riders tightly bunched, Marquez made a move on Dovizioso on the back straight on lap four and was rarely threatened for the remainder of the race.
It’s Marquez’s 20th career victory in the premier class and he remains the favourite to claim a third-straight title.
