Mayer’s decision comes just one day after Parramatta Eels boss Scott Seward parted ways with the club following the Eels’ salary cap issues.
Wests have struggled in the two seasons since Meyer took over from Stephen Humphreys in 2013, finishing last in 2013 and 13th in 2014.
The Tigers have lost six of their past eight games this season and currently sit in 11th.
Club chair Marino Go thanked Mayer for his time at the club, but said the Tigers would now go in a different direction.
“Wests Tigers would like to thank Grant for his diligence and dedication to the club,” Go said in a club statement.
“We look to take the next step in our evolution and a fresh approach to take us to the next level as a professional, world-class sporting organisation.
“Wests Tigers is looking forward to achieving the goals that are critical for a successful future for the club, both on-field and off-field.”
Head of football Phil Moss will become acting CEO until a replacement is found.
