The 39-year-old has dominated the division for many years, and although he is expected to overcome his latest rival, Jennings does represent a real threat to his titles given his unbeaten professional run so far.
The American has youth, athleticism and speed in his favour, but what he lacks is experience and so it remains to be seen whether or not he can put himself in a position to catch the champion clean enough to produce one of the biggest upsets the sport will have seen.
With the likes of Deontay Wilder and Tyson Fury waiting in the wings for a shot at the Ukrainian, Jennings will be hoping to ruin the party for them and become the new heavyweight king at the Garden.
However, the pair have already seen the limelight stolen from them after Wilder got into a heated confrontation with Shannon Briggs at the weigh-in on Friday.
Briggs has stalked Klitschko for well over a year now as he continues to try and goad him into a fight, but his attempts have fallen on deaf ears and so he arguably looked to switch his attention to the WBC champion with a summer bout now a possibility.
However, Wilder has made no secret of his desire to go on and unify the division, and so along with many other star names in the weight class, he will have a close eye on proceedings in New York on Saturday to see whether or not Klitschko can hold on to his belts.
The veteran fighter is delighted to be back fighting in the USA after seven years away, and that excitement could well ensure that he has not underestimated his opponent and he will look to put on a show on his return to HBO and prove that he still has plenty left in the tank yet.
Prediction: Klitschko stoppage in rounds 6-9.
